Our mission is to provide researchers with a single facility where they can access a comprehensive range of resources and services for gene transfer purposes. The Core constructs purifies, and titers custom recombinant adenovirus, adeno-associated virus, and lentivirus. The core is administered via the Department of Physiology and Biophysics and is physically located in COMRB2082. Adenovirus:
Track 3: Amplification of adenovirus from customer's virus, 2 ~ 4 weeks time, volume: 1-2 ml, Titer: >1X10^10 pfu/ml.
Track 4: Amplification of adenovirus from customer's virus, 4 ~ 8 weeks time, volume: 1-2 ml, Titer: >1X10^11 pfu/ml.
Generation of adenovirus – individual investigators construct the shuttle vector, verify, and supply the DNA to the core. Core does recombination and virus generation.
Adeno-Associated Virus (AAV): Core has AAV serotypes (1, 2, 8, and 9) currently.
Track 2: Investigator provides sufficient verified AAV shuttle DNA. 1-2 months time, ~0.5ml, Titer: >1E13 vector genomes (Vg/ml)
Lenti-/Retro-virus:
Virus titration
The viral vector core can measure your virus titer ( 1-2 weeks time).
Subcloning:
Core does subclone for virus vectors with knockdown, knockout, overexpression, tagging, mutation, etc.
For more specific services available (promoter options, mutagenesis, fusion protein construction, etc), please contact Dr. Youn.
Users are strongly encouraged to contact Dr. Youn in the planning process of a project.
Please note, Investigators from Northwestern University and the University of Chicago are provided internal rates as all UIC users.
All other users can access services but, at either external academic or external non-academic rate.

Homologue recombination and adenovirus synthesis |
The customer provides a shuttle vector around 1 ug and their plasmid map. The Core does the homolog recombination from the customer's shuttle vector and virus generation (2~3 weeks). To amplify an adenovirus, the customers need to request other amplification services (Track 1 to 4), depending on their purpose. The plasmid amplification and the cloning of the shuttle vector are separated from this service. |

The customer provides ~20ug of expression vector per virus. If the customer don't have enuough expression vector, it will be added to the service charges ($30, transformation and mini-prep). |

Lenti-/Retro-virus track 3 |
Amplification of Lenti-/Retro-virus from customer's vector with concentration, ~ 2 week time, volume: 0.1 ~ 1ml, Titer: >1X10^8 TU/ml. If the customer doesn't provide enough expression vector (300ug), it will be added to the service charges ($70, transformation and midi-prep). The current concentration service is a PEG-based procedure. If the customer wants to use ultracentrifuge-based concentration without PEG precipitation, we will add the additional Ultracentrifuge service charge (Internal:$60 and exteranl:$70). |
